Research on Degradable Polyurethane Bone Adhesive

    • Bone adhesive provides a simple and convenient method for the treatment of bone fractures. It is not necessary to remove of the metal implants. Polyurethane(PU) is a kind of material with good biocompatibility, physical and mechanical properties, and has a wide range of applications in medical fields. A two-component polyurethane adhesive system was designed and its feasibility as a bone adhesive by in vitro experiments was investigated. Briefly, polyester triol was successfully synthesized with ε-caprolactone, glycolide and D, L-lactide as the monomers, glycerol as the initiator and stannous octoate as the catalyst. The synthesized polyurethane triol was then reacted with isocyanate terminated prepolymer to form the polyurethane adhesive. The chemical structure of the polyester triol and PU adhesive were characterized by nuclear magnetic resonance (1H-NMR) spectroscopy and Fourier transform infrared (FT-IR) spectroscopy. Surface morphology, mechanical properties and thermodynamic properties of polyurethane adhesives were evaluated by scanning electron microscopy (SEM), universal tensile testing machine, and differential scanning calorimetry (DSC). The degradation rate of PU adhesive was tested by placing PU disks in a 10 mL glass vial filled with 0.1 mol/L phosphate buffer saline (PBS) solution at pH 7.4. Effects of polyestertriol molecular weight and the incorporation filler on the properties of PU adhesives were explored. The results showed that the PU-6C400-F group presented good compressive performance with a bonding strength of 0.93 MPa, and with a compressive strength and modulus of 12.5 MPa and 128.0 MPa, respectively. The PU adhesive could be a good candidate for bone gluing applications.
