    高分子微球偶联固定培美曲塞二钠

    Covalent Immobilization of Pemetrexed Disodium on Polymer Microspheres

    • 摘要: 在无皂乳液聚合制备表面带环氧基的聚(苯乙烯甲基丙烯酸环氧丙酯)(PSG)乳液微球的基础上,采用1,6己二胺对PSG微球进行表面修饰,获得了氨基封端、表面清洁的PSG1,6己二胺(PSGN)微球。通过PSGN微球表面的端氨基反应性偶联固定培美曲塞二钠药物分子,考察了药物分子偶联固定反应的影响因素。结果表明:培美曲塞二钠药物在PSGN微球表面的固定接枝量主要取决于PSGN微球的粒径大小和表面活泼端氨基的质量浓度及分布。

       

      Abstract: Poly(styrenecoglycidyl methacrylate) (PSG) latex microspheres with epoxy groups on their surface are prepared by soapfree emulsion polymerization. PSG microspheres are surface grafted with 1, 6hexanediamine to prepare PSG1, 6hexanediamine(PSGN) microspheres. The pemetrexed disodium drug molecules are then covalently immobilized on the surface of the PSGN particles through the reaction with end amino groups. The factors of drug molecules covalent immobilization are studied. Results show that the pemetrexed disodium molecules covalent immobilization on the PSGN microsphere surface is mainly dependent upon the particle size and the concentration and distribution of active amino groups.
